Eukaryotic cell lysate
Lysis buffer: 10 mM Tris, pH 8.5, 5 mM EDTA, 200 mM NaCl, 0.2% SDS
DigestTip digestion time: 1 minute
Lysis buffer: 10 mM Tris, pH 8.5, 5 mM EDTA, 200 mM NaCl, 0.2% SDS
DigestTip digestion time: 1 minute
Lane A): Protein extract from 10E6 ?TC-1 cells in 1 ml of lysis buffer.
Lane B) Protein extract digested with proteinase K at a concentration of 140 ?g/ml at 60°C for 1 hour.
Lane C): Protein extract digested with DigesTip Proteinase K at room temperature for 1 minute.
Lane D): Molecular Weight Standard.

Hybridization
Lysis buffer: 50 mM tris pH 7.5, 5 mM EDTA, 200 mM NaCl, 1% SDS.
Southern blot analysis. Murine stem cells were solubilised with lysis buffer and digested with proteinase K or DigesTip proteinase K. After digestion, genomic DNA was extracted with the phenol/chloroform method, digested with EcoRI and separated on agarose gel, then transferred to a nylon filter and hybridized with 32P–specific probe.
Lysis buffer: 50 mM tris pH 7.5, 5 mM EDTA, 200 mM NaCl, 1% SDS.
Southern blot analysis. Murine stem cells were solubilised with lysis buffer and digested with proteinase K or DigesTip proteinase K. After digestion, genomic DNA was extracted with the phenol/chloroform method, digested with EcoRI and separated on agarose gel, then transferred to a nylon filter and hybridized with 32P–specific probe.
Lane A): Sample digested overnight with Proteinase K, 300 ?g/ml, at 55°C.
Lane B) Sample digested with DigesTip Proteinase K for 1 minute at room temperature.
Lane C): Negative control.
